In vitro transcription is a new technology that uses DNA templates to synthesize mRNA in vitro. Stemirna is one of the first companies in China to establish a full IVT mRNA platform. Structure of mRNA
Template optimization: the IVT mRNA template is related to the stability and translational efficiency of mRNA in the cells.
5' untranslated region (5' UTR) and 3' untranslated region (3' UTR) are untranslated regions flanking the coding region. The untranslated regions contain the regulatory sequences.
PolyA tail is a sequence of repeated adenosines located outside the 3' untranslated region.
Codon optimization in coding sequence: the combination of codons in the coding region can be optimized to increase translational efficiency without changing the amino acid sequence encoded.
Modified nucleotides: modified nucleotides can effectively help IVT mRNA avoid attacks by the immune system, therefore increasing translational efficiency.
